I don't think Diamond Tours exists anymore, but as a local to Stratford-upon-Avon I'm happy to help! Your itinerary sounds great and very do-able. If you will have a car then it's easy to get between the places you mention by road, but if not then they're also pretty well connected by train - though this may take a bit longer, you can relax and watch the countryside roll by! From London you can take a train to Oxford and take a tour of Bleinheim Palace from there. To get to Stratford-upon-Avon from Oxford you'll need to take a train to Banbury and change there. There are plenty of options for town walking tours or sightseeing bus tours in both Oxford and Stratford, and you can take a day tour of the Cotswolds from either town too. To get to Southampton you can travel from Stratford-upon-Avon to Leamington Spa station, and change there to take a direct train to Southampton (though check if you're travelling on a weekend as sometimes the timetables are different or engineering works mean there is a replacement bus service). The last leg of that journey takes just over 2 hours.
