Sorry for delayed response. If you are staying overnight at Labuan Bajo then you can start at about 8 am and return by 4 pm. The travel time by boat to Rinca island is only 2.5 hours. One hour in the park. Add one hour to park the boat and get out of berth. So in all you need 7 hours for boat. Barbour to airport transfer is 20 min. So it is quite possible. I can connect you with my tour operator for this.
However if you intend to land at Labuan Bajo in morning, you can't reach Barbour before 10 am then it becomes risky for return flight.
